The enzymatic oxidation of pyridoxine and pyridoxamine phosphates.

چکیده
Extracts of rabbit liver and brain are known (1, 2) to oxidize pyridoxine 5-phosphate to pyridoxal 5-phosphate much faster than they oxidize free pyridoxine to pyridoxal. Comparison of the two activities during limited purification indicated that different enzymes catalyzed the two reactions (1). Pogell (3) had shown previously that oxidation of pyridoxamine 5-phosphate was catalyzed by an enzyme present in rabbit liver. This enzyme was purified approximately 15-fold and was shown to be distinct from both monoamine oxidase and diamine oxidase. Further investigations of the purification and properties of pyridoxine 5-phosphate oxidase and of pyridoxamine 5-phosphate oxidase, reported herein, lead to the conclusion that the two enzymes are probably identical. In the course of these investigations, a simple photometric method has been developed for the determination of pyridoxal or pyridoxal S-phosphate in enzymatic reaction mixtures.

Transport and metabolism of vitamin B6 in lactic acid bacteria.
Streptococcus faecalis 8043 concentrates extracellular [3H]pyridoxal or [3H]pyridoxamine primarily as the corresponding 5'-phosphates. Accumulation of pyridoxamine requires an exogenous energy source and is inhibited by glycolysis inhibitors. A membrane potential is not required for transport of pyridoxamine, and an artificially generated potential does not drive uptake in this organism. Based ...
متن کاملTHE VITAMIN Bg GROUP
Evidence for the production in viva from pyridoxine of compounds with increased activity for certain lactic acid bacteria (1) and for the production of similar compounds by amination or oxidation procedures in vitro (2, 3) has been reported. Only a limited number of structures for the compounds resulting from amination or oxidation appeared possible; synthesis (4, 5) of certain of these resulte...
متن کاملAll-enzymatic HPLC method for determination of individual and total contents of vitamin B6 in foods
BACKGROUND There is a need for a reliable and accurate method for quantification of each of the seven individual vitamin B(6) compounds including pyridoxine-β-glucoside in foods. OBJECTIVE To determine pyridoxal (PL), pyridoxamine (PM), pyridoxine (PN), pyridoxal 5'-phosphate (PLP), pyridoxamine 5'-phosphate (PMP), pyridoxine 5'-phosphate (PNP), and pyridoxine-β-glucoside (PNG) in foods. DE...
متن کاملB-vitamin Nutrition in Dairy Cows
The B-vitamins include thiamin (B1), riboflavin (B2), niacin (B3), pantothenic acid (B5), the B6 complex (pyridoxal, pyridoxamine, pyridoxine), biotin (B8 or Vitamin H), folic acid (B9), and B12. Each of the B-vitamins plays a key role as either an enzymatic cofactor or metabolic constituent in many facets of intermediary metabolism. For example, about half of the propionate that reaches the ru...
متن کاملPurification and characterization of vitamin B6-phosphate phosphatase from human erythrocytes.
Human erythrocytes rapidly convert vitamin B6 to pyridoxal-P and contain soluble phosphatase activity which dephosphorylates pyridoxal-P at a pH optimum of 6-6.5. This phosphatase was purified 51,000-fold with a yield of 39% by ammonium sulfate precipitation and chromatography on DEAE-Sepharose, Sephacryl S-200, hydroxylapatite, and reactive yellow 86-agarose. Sephacryl S-200 chromatography and...
